Electrospun polyurethaan / poly (ɛ-caprolacton) nanovezels bevorderden de hechting en voortgang van menselijke endotheelcellen in statische en dynamische traditiesituaties

• Bij dit onderzoek werd het angiogene vermogen van menselijke endotheelcellen bestudeerd na 72 uur op de vloer van polyurethaan-polycaprolacton (PU / PCL) steigers te zijn geplateerd. Bij dit onderzoek zijn cellen ingedeeld in 5 totaal verschillende teams, samen met PU, PU / PCL (2: 1), PU / PCL (1: 1); PU / PCL (1: 2); en PCL.

• Uit informatie bleek dat de samenstelling van PU / PCL (2: 1) de volgende modulus en breekpunt had in vergelijking met de tegenovergestelde teams (p <0,05). In vergelijking met de tegenovergestelde teams had de PU / PCL-steiger met een molaire verhouding van twee: 1 een kleinere contacthoek θ en een betere trekspanning (p <0,05).

• De impliciete meting van de PU-nanovezels werd verlaagd na toevoeging van PCL (p <0,05). Voornamelijk gebaseerd op onze informatie, veroorzaakte de traditie van endotheelcellen op de vloer van PU / PCL (2: 1) geen nitrosatieve stress en cytotoxische resultaten onder statische situaties in vergelijking met cellen die op een traditionele plastic vloer waren geplateerd (p> 0,05) .

• Voornamelijk gebaseerd op informatie uit de statische situatie, hebben we een buisvormige PU / PCL (2: 1) assemblage vervaardigd voor zesdaagse dynamische celtraditie in lusluchtliftbioreactoren. Scanning-elektronenmicroscopie bevestigde de hechting van endotheelcellen aan de luminale vloer van de PU / PCL-steiger. Cellen zijn afgevlakt en uitgelijnd onder de traditionele mediumbeweging. Immunofluorescentiebeeldvorming bevestigde de hechting van cellen aan de luminale vloer, aangegeven door blauwe kernen op de luminale vloer.

• Deze informatie toonde aan dat het gebruik van PU / PCL-substraat de lichaamsbeweging van endotheelcellen kan stimuleren onder statische en dynamische situaties.

In het laatste decennium heeft de driedimensionale (3D) celtraditie-knowhow een aantal nieuwsgierigheid gekregen die kan worden toegeschreven aan het potentieel om de in vivo-groep en de micro-omgeving van in vitro gekweekte kankercellen beter te recapituleren.

In het bijzonder hebben 3D-tumormodellen een aantal totaal verschillende eigenschappen aangetoond in tegenstelling tot conventionele tweedimensionale (2D) culturen en hebben ze gezorgd voor een opvallende hyperlink tussen de laatste en dierproeven.
